The Effect of the Endothelial Cell culture Mixed with Vascular Endothelial Growth Factor(VEGF) and Cyclosporin A (CsA) in Rabit Aorta

Abstract
The intimal hyperplasia is one of the main causes of early failure in arterial bypass graft. The absence of injury of vascular endothelial cell lining causes platelet aggregation, thrombi formation and secretion of platelet drived growth facto
and
ultimately promote the proliferation of the smooth muscle cell and the intimal hyperplasia. The purpose of this study was to investigate the effect of CsA and VEGF on the growth and inhibition of the endothelial cells(EC).
We got EC from abdominal aorta of 10 adult rabbits and then cultured in the culture Media 199 up to 9 days. As a control group, EC were lutured in Bovine calf serum. As experimental groups, we added 1% VEGF to a control group as a grou I. 1% VEGF
and
CsA 0.5, 5, 50, 500 ng/ml(non-toxic level of clinical cosage) respectively as a group II, 1% VEGF and CsA 0.5, 5, 50, 500§¶/ml respectively as a group¥².
We measured growth rate of the EC with the MTT assay.
As compred with te control group and group I, the growth rate of the latter was markedly increased at the fifth day of culture. As compared with the grwoth rate of the group I and group II, both two groups ahd no difference. So the inhibition of
EC
groupwth was suppressed at the level of 50§¶/ml and we could not be observed at the non-toxic level of CsA. In the group ¥², Ec growth was suppressed at the level of 50§¶/ml and we could not observe the EC growth iself at the level of 500§¶/ml.
In conclusion, VEGF was essential factor of EC culture and CsA below dosage of 50§¶(500ng)/ml could not suppress the EC growth.
We can expect long-term vascular graft patency whe we use the proper dosage of CsA that can suppress the intimal hyperplasia without any influence ot the growth of the EC.
